- Pioneera Biosciences (Pioneera Biomokslai UAB, Vilnius, Lithuania) is an immuno-oncology company engineering cell therapies that are impossible to escape. Its Discovery 4.0 platform treats antigen escape - the leading engineerable cause of relapse after CAR-T and CAR-NK therapy - as a first-class design objective: a mutation-epitope risk engine scores Clinical Escape Likelihood per residue before anything is built, and full-stack design (epitope foundation encoder, docking and sequence intelligence generating 5,000 synthetic binders per antigen, multivalent construct generator, effector-cell matching across 8 subtypes) is calibrated by ~100 proprietary in vitro co-incubation experiments. In head-to-head models the tri-specific lead shows 0.09% effective escape probability versus 9-19% for monospecific controls. The pipeline runs one engine, two programs: PNR-212, an allogeneic mRNA dual-CAR-NK targeting CD19 for autoimmune disease and B-cell malignancies (2.2x median survival, complete tumour regression in 89% of treated mice), and PNR-101, an autologous anti-BCMA dual-CAR-T for oncology (1.7x survival, 83% complete regression). Both are IND-enabling, with IND submission planned Q1-Q2 2027. The platform takes a target to clinical candidate in ~6 months for under $1M per program.

- Most cell-therapy patients respond. Most then relapse. 30-60% of relapses after CAR-T / CAR-NK involve antigen escape, and 42% of relapsed CAR-T patients relapse CD19-negative. The field optimised affinity, potency and persistence - it never optimised against escape.
